dc.description.abstract The success of the fight against animal brucellosis depends on the effectiveness of diagnostic studies. The scientific interest in studying the species composition of the causative agent of brucellosis is important, since this approach can identify sources and reservoirs of infection, correctly and objectively develop a scientifically based scheme of anti-brucellosis measures, a system of measures aimed at protecting people from infection with brucellosis, as well as differentiation of brucellosis pathogens. In recent years, molecular genetic research methods, in particular, the polymerase chain reaction, have been used to identify, differentiate brucella and confirm laboratory diagnosis. PCR allows in a short time to determine the presence of a specific Brucella DNA sequence in samples of both clinical and field material. The appearance of the PCR method was due to certain achievements of molecular genetics, primarily the decoding of the nucleotide sequence of the genomes of a number of microorganisms. It is impossible not to say that PCR was made possible by the discovery of the unique enzyme taq-DNA polymerase, which is found in bacteria living in geysers. The peculiarity of this polymerase lies in its exceptional heat resistance (withstands heating to boiling point without loss of activity) and high operating temperature (optimum operation - 72 0C)Успех борьбы против бруцеллеза животных зависит от эффективности диагностических исследований. Научный интерес изучения видового состава возбудителя бруцеллеза важен, так как при этом подходе можно выявить источники и резервуары инфекции, правильно и объективно разрабатывать научно обоснованную схему противобруцеллезных мероприятий, систему мер, направленных на ограждение людей от заражения бруцеллезом, а также дифференциация возбудителей бруцеллеза. В последние годы для идентификации, дифференциации бруцелл и лабораторного подтверждения диагноза используют молекулярно-генетические методы исследования, в частности полимеразную цепную реакцию. ПЦР позволяет в короткие сроки определить наличие специфической последовательности ДНК бруцелл в пробах как клинического, так и полевого материала. Появление метода ПЦР было обусловлено определенными достижениями молекулярной генетики, прежде всего расшифровкой нуклеотидной последовательности геномов ряда микроорганизмов. Нельзя не сказать, что ПЦР стала возможной благодаря открытию уникального фермента taq-ДНК-полимеразы, содержащегося у бактерий, обитающих в гейзерах. Особенность этой полимеразы заключается в ее исключительной термостойкости (выдерживает нагревание до температуры кипения без потери активности) и высокой рабочей температуре (оптимум работы - 720С) ru
